SCO-101 is advanced as a combination partner across six oncology programmes led by CORIST, a Phase IIa study in metastatic colorectal cancer.
Phase I–IIa · SCO-101 + FOLFIRI
The CORIST Phase I–IIa study investigates SCO-101 in combination with the chemotherapy regimen FOLFIRI in patients with irinotecan-resistant metastatic colorectal cancer (mCRC).
Patients enrolled in CORIST have failed all prior standard chemotherapy and have entered a palliative stage of their disease — with little hope of either a cure or of extending life further.
The first part established a safe dose — the maximum tolerated dose — of SCO-101 when given together with FOLFIRI. It was successfully completed in June 2021.
Based on findings in Part 1, the second part enrolled only patients with RAS wild-type tumours, focusing on safety, tolerability and efficacy of SCO-101 on a schedule combining SCO-101 and FOLFIRI over seven days.
Part 3 expanded the trial by adding a new schedule combining SCO-101 and FOLFIRI over four days, in patients with both RAS wild-type and RAS mutated irinotecan-resistant tumours.
